Stem Cell Bioengineering with Bioportides: Inhibition of Planarian Head Regeneration with Peptide Mimetics of Eyes Absent Proteins

Djeya1 (RKLAFRYRRIKELYNSYR) is a very effective cell penetrating peptide (CPP) that mimics the α5 helix of highly conserved Eya domain (ED) eyes absent (Eya) proteins. The objective this study was to bioengineer analogues that, following translocation into planarian tissues, would reduce ability neoblasts (totipotent stem cells) and their progeny regenerate anterior pole in decapitated S. mediterranea. As strategy increase propensity for formation, molecular bioengineering achieved by mono-substitution helicogenic aminoisobutyric acid (Aib) at three species-variable sites: 10, 13, 16. CD analyses indicated helical, Aib-substitution had subtle influences upon secondary structures bioengineered analogues. Aib-substituted are efficient CPPs, devoid influence viability or proliferation. All peptides migration PC-3 cells, prostate cancer line expresses high concentrations Eya. Two peptides, [Aib13]Djeya1 [Aib16]Djeya1, bioportides which delay head regeneration. only population capable division planaria, these data indicate bioportide technologies could be utilised directly manipulate other cells situ, thus negating any requirement genetic manipulation.
